Try to lock these in whenever you get these for your damage dealers:
Eldritch slayer
Eldritch hater
Beast slayer
Beast hater
Most of the most difficult bosses are either Eldritch or Beast. (or even Eldritch/Beast)

For support characters such as Vestals, I like to lock in these:
Hard Nogging
Quick Reflexes
I don't want my vestals to die and I certainly don't want them stunned. I want all the spd in the world to them as well.

For all characters:
Thick Blooded
Most of the time your heroes will die due to bleed or Blight while at death's door so adding resistance to those conditions is great

Similarly, I consider Thin blooded, Anemic, Shocker and Fear of Eldritch/Beast some of the worst quirks.

The "best" negative quirks are ones that limit your use of stress healing facilities since those do not affect your in mission performance at all. also I dont mind winded on my non-healer heroes because I want my healers to act first anyways.

Hope this helps.

My favorite quirks on any character are Steady and Natural Swing. I'm also a fan of Photomania (and Warrior of Light for the damage dealers). Clotter is useful almost everywhere, and Thick Blooded is not far behind. Hard Skinned has been growing on me, as well.

Sometimes I'll build around a region. The Adventurer and Explorer quirks won't help you in the final dungeon but they're quite good when they're active. For those characters I then prioritize quirks that are especially good in the region, like Slayer for the prominent enemy group and resistance quirks against the region's common status effects.

Hard Noggin is good for healers and guardians. Quick Reflexes shines on stunners, healers, and buffers.

Slugger and Precise Striker are notable for melee damage specialists, while Unerring and Eagle Eye are worthwhile on ranged damage dealers.

Evasive is one I'll lock in for Jester because his base dodge is so high, and for Antiquarian because I favor her dodge-buffing skill in later levels.

The Slayer quirks are great; accuracy never stops being important and crits heal stress. The best is probably Eldritch because of how common it is, followed by Beast because of how many beasts are extra evasive.

There is no fast to level up, you just need to grind the dungeons and beat the bosses, if you will encounter the Shambler kill it a few times it drops an item named "Ancestor's Portrait" gives 50% xp and +10% stress, you can use it on a character to level up it faster.
